RESUMO

A quantitative method based on quick, easy, cheap, effective, rugged, and safe technique (QuEChERS) sample extraction and ultra-performance liquid chromatography/tandem mass spectrometry (UHPLC-MS/MS) was evolved for the determination of 47 pesticide residues in fresh Mazafati date fruits from Bam City of Kerman Province, Iran. The recoveries for selected pesticides ranged from 88 to 110% with a relative standard deviation (RSD) of less than 20% at concentrations of 0.05 and 0.1 mg kg-1. The proposed method had a linear range from the limit of quantification (LOQ) to 1.00 mg kg-1, and the LOQ of the 47 pesticides was ≤ 0.005 mg kg-1. The coefficients of determination (R2) were more than 0.99. This technique was used on 12 fresh date fruits samples, three water samples, and three soil samples with three replications per sample. Forty-seven pesticide were detected collectively, but only diazinon was detected in the date fruit samples. The mean value of diazinon residues was 0.037 mg kg-1, and the concentration of diazinon in most samples was below the national maximum residue limit (MRL) for date fruit (0.05 mg kg-1). Among the pesticides measured, diazinon residues were also detected in the water samples, but not in the soil samples. The dietary intake assessment showed no health risk to humans from the consumption of fresh date fruit concerning the pesticides investigated.

Detection of pesticide residues in soil samples was conducted using UHPLC-MS/MS. Non-dietary health risk assessment was conducted using calculate chronic daily intake (CDI) from ingestion, inhalation and dermal contact pathways and following non-carcinogenic and carcinogenic risks in the adults and adolescent. The rank order of pesticide in soil based on their concentration was malathion (0.082 mg kg-1)> cyproconazole (0.019 mg kg-1)> propargite (0.018 mg kg-1)> butachlor (0.016 mg kg-1) > chlorpyrifos (0.0067 mg kg-1)> diazinon (0.0014 mg kg-1)> imidacloprid (0.0007 mg kg-1). Hazard index (HI) values obtained of exposure to pesticides in soil in adults and adolescent were 0.0012 and 0.0035, respectively. Hence, exposed population are at the acceptable range of non-carcinogenic risk (HI < 1). Cancer risk (CR) values due to propargite in soil via ingestion pathway in adults and adolescent were 2.03E-09 and 2.08E-09, respectively; therefore, carcinogenic risk due to the exposure to pesticide contaminated soil was safe range (CR < 1E-06).

The chronic dietary risk assessment for 34 pesticides suspected of acting as endocrine disrupters in Iran was assessed by comparing TMDI with the Acceptable Daily Intakes (ADI). Of 34 investigated endocrine-disrupting pesticides (EDPs), 6 had TMDI > ADI. In addition, potential non-carcinogenic and carcinogenic health risk assessments were evaluated using Monte Carlo simulation. HQ in wheat was 17.40 and 20.29 in adults and children, respectively. Due to dimethoate residue in wheat, HQ was 2.78, and for fenitrothion residue, 3.22. HI was 21.22 for adults and 24.76 for children in wheat, more than 1. Total Carcinogenic risk (TCR) due to EDPs residues was 6.40 × 10-5 in apples, in citrus fruits 5.97 × 10-5, 3.33 × 10-5 in cucumber, 5.30 × 10-5 in lettuce, in potato was 2.36 × 10-5, in rice was 1.61 × 10-5, 1.78 × 10-5 in tomato, and due to epoxiconazole residues in wheat was 3.18 × 10-5, more than acceptable limit 1.0 × 10-6. End-stage of liver fibrosis as a precancerous state could lead to cirrhosis and hepatocellular carcinoma which liver transplantation is the only effective treatment. Previous studies have indicated that farnesoid X receptor (FXR) agonists, such as obeticholic acid (OCA) protect against hepatic injuries. However, free OCA administration results in side effects in clinical trials that could be alleviated by applying bio carriers such as MSC-derived exosomes (Exo) with the potential to mimic the biological regenerative effect of their parent cells, as proposed in this study. Loading OCA into the Exo was conducted via water bath sonication. Ex vivo bio distribution studies validated the Exo-loaded OCA more permanently accumulated in the liver. Using CCL4-induced liver fibrosis, we proposed whether Exo isolated from human Warton's Jelly mesenchymal stem cells loaded with a minimal dosage of OCA can facilitate liver recovery. Notably, Exo-loaded OCA exerted additive anti-fibrotic efficacy on histopathological features in CCL4-induced fibrotic mice. Compared to baseline, Exo-mediated delivery OCA results in marked improvements in the fibrotic-related indicators as well as serum aspartate aminotransferase (AST) and alanine aminotransferase (ALT) concentrations. Accordingly, the synergistic impact of Exo-loaded OCA as a promising approach is associated with the inactivation of hepatic stellate cells (HSCs), extracellular matrix (ECM) remodeling, and Fxr-Cyp7a1 cascade on CCL4-induced liver fibrosis mice. In conclusion, our data confirmed the additive protective effects of Exo-loaded OCA in fibrotic mice, which suggests a valuable therapeutic strategy to combat liver fibrosis. Furthermore, the use of Exo for accurate drug delivery to the liver tissue can be inspiring.

RESUMO

Endocrine-disrupting chemicals (EDCs) are a growing public health concern worldwide. Consumption of foodstuffs is currently thought to be one of the principal exposure routes to EDCs. However, alternative ways of human exposure are through inhalation of chemicals and dermal contact. These compounds in food products such as canned food, bottled water, dairy products, fish, meat, egg, and vegetables are a ubiquitous concern to the general population. Therefore, understanding EDCs' properties, such as origin, exposure, toxicological impact, and legal aspects are vital to control their release to the environment and food. The present paper provides an overview of the EDCs and their possible disrupting impact on the endocrine system and other organs.

This research is aimed at the analysis of 87 pesticides in 30 fresh pistachio samples prepared from stores in Iran by QuEChERS-ultra-high performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS). The results showed at least one pesticide was in 67% of fresh pistachio samples. Kresoxim methyl residue was detected in 20 samples with average of 0.11 mg kg-1; this average is 2.2 times more than maximum residue limit (MRL). Buprofezin was recognized in five samples with average of 0.17 mg kg-1 was observed with 3.4 times more than MRL; hexaconazole and permethrin were recognized in three samples lower than MRL with an average residue of 0.030 and 0.028 mg kg-1, respectively. In addition, potential non-carcinogenic and carcinogenic health risk assessments were evaluated using probabilistic methods performed with the Monte Carlo simulation algorithm. The order of pesticide ranking based on hazard quotient (HQ) was hexaconazole > buprofezin > permethrin > kresoxim methyl. Total HQ (HI) was 2.0E-4 and for children, 9.0E-4. Hence, it means fresh pistachio consuming maybe not have significant short-term health risks for consumer. Ranking based on cancer risk (CR) was hexaconazole > kresoxim methyl > metalaxyl > permethrin > buprofezin. However, total CR due to pesticide residues was not higher than 1E-6 value (1.09E-9); therefore, consumers were not at significant risk of carcinogenicity in this product.

In this research, walnut shell along with a Ni-metal organic framework (Ni-MOF) was thermally treated to obtain a highly efficient biochar. A µ-solid phase extraction (µ-SPE) setup containing the optimized walnut modified biochar was implemented for the extraction of 20 wheat specified pesticides with a wide range of polarities. The modified biochars were characterized and the role of Ni-MOF on the porosity of the obtained biochars was investigated. After optimizing the extraction parameters, the 50 %-Ni-MOF-walnut shell biochar was selected. Accordingly, the precision for intra-day (2.4-25 %), inter-day (7-28.3 %) and synthesis procedure (4.1-26.7 %) was calculated. The limits of detection and quantification were in the range of 0.01-12.1 µg/L and 0.03-40.5 µg/L, respectively. This method was applied for the analysis of 29 wheat samples. The risk assessment study demonstrates that the contaminated wheat samples cannot cause non-carcinogenic harm to adults but might have an impact on children.

Due to the widespread use and environmental pollution of estrogenic chemicals, the need for screening tests to detect these compounds is felt more than ever. These compounds lead to cell proliferation. Therefore, studies used cell proliferation to evaluate estrogenic compounds was studied in this systematic review. This systematic review was performed with the keywords; DNA proliferation, cell proliferation, estrogenic component, estrogen, food, bioassay, screening, and detection. After initial screening and full text quality assessment, 16 manuscripts were selected and data were extracted. Four cell lines, MCF-7, MDA-MB-231, Ishikawa, and T47D cells were used in the studies. MCF-7 was more sensitive to estrogenic compounds than other lines. Most of the samples studied were plant compounds and mycotoxins and substances that migrate from packaging to food. This screening test is valid and has similar results as others.

Contamination of food with mycotoxins can pose harmful effects on the health of consumers in the long term. Coffee contamination with mycotoxins has become a global concern. This study attempted to meta-analyze the concentration and prevalence of ochratoxin A (OTA) in coffee products and estimate consumers' health risks. The search was conducted among international databases, including Scopus, PubMed, Embase, and Web of Science, for 1 January 2010 to 1 May 2022. The concentration and prevalence of OTA in coffee products were meta-analyzed according to country subgroups. Health risk assessment was conducted based on Margin of Exposures (MOEs) using the Monte Carlo simulation (MCS) technique. The three countries that had the highest Pooled concentration of OTA in coffee were observed in Chile (100.00%), Kuwait (100.00%), and France (100.00%). The overall prevalence of OTA in coffee products was 58.01%, 95% CI (48.37-67.39). The three countries that had the highest concentration of OTA were Philippines (39.55 µg/kg) > Turkey (39.32 µg/kg) > and Panama (21.33 µg/kg). The mean of MOEs in the adult consumers in Panama (9,526) and the Philippines (8,873) was lower than 10,000, while the mean of MOEs in other countries was higher than 10,000. Therefore, monitoring and control plans should be carried out in different countries.

Acrylamide, a food-borne chemical toxicant, has raised global concern in recent decades. It mainly originated from reducing sugar and free amino acid interactions in the carbohydrate-rich foodstuffs heated at high temperatures. Due to the neurotoxicity and carcinogenicity of AA, the mechanism of formation, toxic effects on health, and mitigation strategies, including conventional approaches and innovative technologies, have been of great interest since its discovery in food. Potato products (especially French fries and crisps), coffee, and cereals(bread and biscuit) are renowned contributors to AA's daily intake. The best preventive methods discussed in the literature include time/temperature optimization, blanching, enzymatic treatment, yeast treatment, additives, pulsed electric fields, ultrasound, vacuum roasting, air frying, and irradiation, exhibiting a high efficacy in AA elimination in food products.

Following the EPA recommended method, a chronic diet risk assessment for pesticides was recently performed for adults and children in the Iranian population. The National Theoretical Maximum Daily Intake (NTMDI) for 32 pesticides was computed using the theoretical maximum residue limits (MRLs) of food consumption regulations and data from the comprehensive database of the Iranian Research Institute of Plant Protection (IRIPP). The risk was assessed by comparing TMDI with the acceptable daily intakes (ADI) evaluated by FAO. From 32 investigated pesticides, 10 pesticides had TMDI > 65% of the ADI. Some of these ADI-exceeding compounds (spirodiclofen, abamectin, trifloxystrobin, spiromesifen, fipronil, difenoconazole, tetraconazole) were found in citrus, cucumber, grapes, tomato, and potato as the foods that have played the most roles in the consumption of these pesticides. Furthermore, a probabilistic risk assessment was performed to estimate the contingency of extravagance of the ADI. In the current research, only cyazofamid in potato for children consumers exceeded the 1 of the HQ. However, carcinogenic risk (CR) due to spirodiclofen in citrus fruit and difenoconazole in tomato was higher than the 1E-6 value; therefore, consumers were at considerable carcinogenic risk in these commodities. This scrutiny is essential for improving the activities' risk assessment, regulation, and surveillance.

Fish as a critical food source is responsible for supplying nearly 20% of the average per capita animal protein for 3.2 billion people worldwide. However, oral exposure to potentially toxic elements (PTE) such as mercury (Hg), cadmium (Cd), lead (Pb), and arsenic (As) through marine fish consumption can cause health risks in consumers. This study aimed to collect information among databases (2000-2020), including ScienceDirect, PubMed, SCOPUS, EMBASE, and Google Scholar, regarding the concentration of PTEs in different types of fish from the Persian Gulf. In this regard, thirty articles containing 266 samples were included in the current systematic review. In addition, the non-carcinogenic risk caused by ingestion of PTEs via consumption of fish was calculated. The average PTE concentrations in fish muscles were presented in the following order: As (0.37 mg/kg ww) > Pb (0.20 mg/kg ww) > Cd (0.17 mg/kg ww) > Hg (0.13 mg/kg ww). However, the mean concentration of PTEs was below the defined level by the World Health Organization, and the findings revealed that the calculated average daily dose (ADD) for Cd and Pb in Iranian children and adult consumers was lower than the reference dose (RfD), whereas the ADD of Hg was higher than the RfD for children consumers. The total target hazard quotient for Iranian and children consumers was >1. The excess lifetime cancer risk values for Pb were safe, unlike those for Cd, which were in an unsafe range (>10-4) for consumers.

Aqueous Extract of Acanthophyllum laxiusculum root (AE) exhibited remarkable foaming profile, emulsification properties and antifungal activity due to the presence of high concentration of total saponins. Total phenolic compounds, another main component, accounted for the AE antioxidant activity. Spray drying of AE, as a thermal process, did not affect the foaming indices of A. laxiusculum Spray-dried Extract (SE) and is a recommended alternate for convenient application in food industry. Addition of SE to the malt extract at accepted levels of total saponin daily intake (below 50 mg/kg) showed positive attribution of malt and carbonation on the foaming quality of SE. Meanwhile, antioxidant activity of commercial malt beverage was enhanced by addition of phenolic compounds containing SE. The 20-60% antifungal inhibitory ratio of the SE developed here is within the applied range of total saponin that supports its application to inhibit the growth of Saccharomyces cerevisiae after malt beverage production. The bitter taste of SE was not sensed in malt beverage at 30 mg/kg total saponin content and suggested A. laxiusculum root extract as a natural additive in malt industry.

In this study, 30 pesticide residues in 45 fresh-eating cucumber, tomato, and okra fruit samples collected from the Khuzestan province as the main agricultural products in Iran using the QuEChERS extraction method were analyzed. In addition, noncarcinogen and carcinogen health risk assessments were evaluated. Results indicated that 93% of cucumber samples had at least one pesticide, of course, less than the maximum residue limit (MRL). All tomato and okra fruit samples were contaminated by diazinon. All pesticides detected in tomato samples were below national MRL except for thiamethoxam in four samples. In okra fruit samples, all detected diazinon and malathion, but only tebuconazole fungicide exceeded MRL. In addition, the hazard index (HI) was 0.23 and 1.06 in cucumber samples, 0.33 and 1.51 in tomato samples, and 5.5E-03 and 0.025 in okra fruit samples in adults and children, respectively. The use of cucumber and tomato may have notable risks in the short term in children group age. Ranking based on total CR was 1.2E-05 in tomato, 7.7E-06 for cucumber, and in okra 9.1E-11 because of the difenoconazole residue. However, significant carcinogenic risk threatens cucumber and tomato consumers.

Raw, pasteurized, powdered cow milk and human milk samples from Tehran, Iran were investigated for the residual of fifty pesticides with the aid of gas chromatography coupled with electron capture detector (GC-ECD) and mass detector for confirmation; and ultra-high performance liquid chromatography-tandem mass spectroscopy (UHPLC-MS/MS). No pesticide residue was determined in more than 91% of examined samples. However, the dimethoate residue was detected in 3 raw milk samples in levels higher than EU recommended MRL. While in 3 human milk samples, organochlorine pesticides residue, p,p'-DDT, and p,p'-DDD was recognized below MRLs, only in 1 human sample residue of p,p'-DDE was more than CODEX recommended MRL. HI in adults and children were 0.72 and 3.55, respectively. However, the health risk assessment based on HI demonstrated that adult consumers are not at considerable risk. The HI, higher than 1 in children, confirms the risks raised due to ingestion of organochlorine (OCP) and organophosphorus (OPP) pesticides via milk consumption. In addition, no carcinogenic risk to milk consumers was calculated. Therefore, implementation of good farming practices on farms, improving the knowledge and consciousness of pesticide users, use other safe methods for pest control such as biotechnology-based, and use a rational program for application of pesticides, continuous monitoring of pesticides in crops, and strict government regulations on pesticide residues in food are recommended.

Pesticide residues in fruits lead to serious public health and environmental problems. This study was undertaken to analyze 57 pesticides residues in 60 commercially available ready-to-eat packed raisins samples from Iran's markets using Quick, Easy, Cheap, Effective, Rugged, and Safe (QuEChERS) approach along with acetonitrile for the extraction, surface adsorbents for clean-up procedure, following with an ultra-high performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS). Moreover, the probabilistic health risk assessment includes non-carcinogenic and carcinogenic risk were estimated by hazard quotient (HQ), Hazard Index (HI) and cancer risk (CR) using the Monte Carlo Simulation (MCS) method. The respective numbers attributed to LOD and LOQ were 0.001-0.03 mg kg-1 and 0.005-0.05 mg kg-1. Results showed that 23% of samples were contaminated by at least one pesticide according to national MRLs. The rank order of pesticides based on HQ was carbendazim > acetamiprid > thiodicarb > iprodione for raisins with percentile 95% benchmark. The HI amount in the adult's age group was 0.001, and in the children's age group was obtained to be 0.006. According to the risk assessment results (HQ and HI < 1 in both age groups), it can be acknowledged that consumption of raisins may not pose remarkable health hazards in short term exposures. The rank order of pesticides based on CR was thiodicarb > iprodion. But total CR (TCR) was lower than 1E-6 value (7.6E-8), so consumers were not at considerable carcinogenic risk in this commodity. Acetamiprid and carbendazim does not pose a cancer risk to humans. Therefore, a dietary exposure assessment for the purpose of assessing cancer risk is unnecessary.

West Azerbaijan, especially the city of Urmia, is the center of Iranian apple and grape production hence the importance of this, residues of 85 pesticides in these products were investigated using modified QuEChERS extraction followed by UHPLC-MS/MS technique. Residues of 17 different pesticides detected in some apple samples. In grape sample only 7 pesticides detected. The levels of the residues found in all apple and grape samples were below the maximum residue levels (MRLs) of Iran, except for iprodione. Health risk assessment associated with pesticide residues in apples and grapes were estimated by hazard quotient (HQ) and hazard index (HI), which indicated that the HI value was lower than 1 in adults and children due to apple and consumption. HI in adults and children were 0.012 and 0.054 in apple; 0.001 and 0.003 in grape samples, respectively. Although the health risk assessment showed that the consumers are not at considerable risk but due to pesticide residue, implement control plans to manage the proper application of this pesticide, or replace it with safer alternatives in apple and grape is required.

This research aims to analyze 56 pesticide residues in 100 green-house cucumber and 150 cantaloupe and melon samples collected from markets in Iran by Quick, Easy, Cheap, Effective, Rugged, and Safe (QuEChERS) extraction method based on analysis with ultra-high performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS). In addition, non-carcinogenic and carcinogenic probabilistic health risk assessments were evaluated by Hazard Quotient (HQ), Hazard Index (HI), and Carcinogenic Risk (CR) based on the Monte Carlo Simulation (MCS) method. According to Iranian regulation, 18% of cucumber and 22% of cantaloupe and melon samples were contaminated by at least one pesticide. Arrange pesticides based on HQ ranking was diazinon > thiacloprid > imidacloprid tebuconazole for cucumbers and chlorpyrifos > thiacloprid > imidacloprid > metalaxyl for cantaloupes and melons. In addition, HI for adults was 0.067 in cucumber and 0.24 in cantaloupe and melon; and for children, 0.30 in cucumber and 0.60 in cantaloupe and melon. Arrange pesticides based on CR ranking was thiacloprid > imidacloprid > metalaxyl > chlorpyrifos > tebuconazole > diazinon. However, CR was more than 1.0E-6 value due to thiacloprid, so consumers were at considerable carcinogenic risk in these commodities.

Biomass, originates from plant- or animal-based materials with a huge potential to be reused. Here we report a simple, rapid and inexpensive method for preparation of modified biochars derived from the banana peel followed by their applications in pipette-tip micro solid-phase extraction (PT-µSPE). Due to the contribution of various effective parameters on modification of banana peel biochars (BPBs), Taguchi design was used to optimize activation temperature, activation repetition, treatment material and impregnation ratio. Efficiency of the prepared BPBs were studied by extraction of twelve various pesticides, as model analytes with an extended range of log P values (1.4-5.7), followed by their determination using ultra-high performance liquid chromatography-tandem mass spectrometry. After finding the most suitable BPB, the affecting parameters on the PT-µSPE performance were optimized. Accordingly, the LOD values of 0.03-10 µg L-1, linear dynamic range of 0.1-200 µg L-1 and a range of RSD values of 5.3-19% were obtained. Eventually, five fruiting vegetables were analyzed and screened for their possible contaminations. Among the tested pesticides, chlorpyrifos, diazinon, malathion, phosalone, propargite and thiophanate-methyl were detected in eggplant, sweet pepper, zucchini and tomato.

The validation of an analytical procedure based on the quick, easy, cheap, effective, rugged, and safe (QuEChERS) approach is presented for multiresidue analysis of pesticides in dates by liquid chromatography-tandem mass spectrometry (UHPLC-MS2). The proposed methodology was applied for simultaneous quantification of 16 pesticides in 50 different date fruits. Method validation was performed regarding accuracy, precision, LDR, LOD, and LOQ, as well as matrix effects. Results of validation were satisfactory, with recoveries higher than 80% for 75% of the samples for 100- and 500- µg L-1 spike levels. Evaluation of the matrix effect revealed that for 81% of the samples, a slight matrix effect was observed. Residues in 92% of the real samples were found below national MRLs. Afterward, hazard quotient (HQ) and total hazard quotient (THQ) of human health risk assessment of pesticides was estimated using a probabilistic approach based on the Monte Carlo (MC) algorithm. Total hazard quotient (THQ) in adults based on the consumption of dates in total samples was estimated to be 7.8% and 36.7% for adults and children, respectively. Since the studied pesticides are registered in the country and are the most widely used pesticides on dates, the occurrence of other pesticide residues seems to be unlikely. Consequently, the applied health risk assessment on Iranian date fruit samples showed that the HQ for adults and children populations indicates no risk to human health.
